Rechecking the rulebook it says "Dens cannot be built on a hex with an enemy Pack Wolf."
Just for the reference, an action selection is still preserved in the game log even after undoing, so it's possible to obtain the required move number when necessary.

I want to build a den on a desert space, it is occupied by an opponent's pack wolf, I'm not given the option to do so.
According to rule book, page 10, under "Building Dens - special rules:", it says "Dens can be made on a hex with an enemy Pack Wolf."
